This gene encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. The protein has a neuregulin-binding domain but lacks an active kinase domain, so it can bind the ligand but cannot transmit the signal. However, it forms heterodimers with other EGFR family members with kinase activity, activating pathways that lead to cell proliferation or differentiation. Gene amplification or overexpression of this protein has been observed in cancers like prostate, bladder, and breast tumors. Alternate splice variants have been characterized, including one isoform that lacks the intermembrane region and is secreted outside the cell, where it modulates the activity of the membrane-bound form. Other splice variants have been reported but are not fully characterized.

Myosin phosphatase target subunit 1 (MBS) is a key component of myosin phosphatase, which regulates actin-myosin interactions downstream of the Rho GTPase. RhoA, when active in its GTP-bound form, interacts with MBS to control myosin light chain (MLC) phosphorylation, influencing smooth muscle contraction and nonmuscle cell movement. Rho-kinase, activated by GTP-bound RhoA, phosphorylates and inactivates MBS, reducing myosin phosphatase activity. Overexpression of RhoA or activated RhoA increases MLC phosphorylation. Several transcript variants encoding different isoforms of this gene have been identified.
